Overview

Peripheral Blood Mononuclear Cells (PBMCs) are a heterogeneous population of primary human immune cells isolated from peripheral blood. They consist primarily of lymphocytes and monocytes and are among the most widely used primary cell preparations for investigating immune responses, inflammation, infectious diseases, cancer immunology, and biomarker discovery.

Unlike immortalized immune cell lines, PBMCs closely reflect the physiological immune status of the donor, making them an indispensable model for translational and clinical immunology research.


Origin

  • Isolated from human peripheral blood.
  • Commonly obtained from whole blood or buffy coat preparations.
  • Routinely separated using Ficoll density-gradient centrifugation.
  • Widely used in basic, translational, and clinical immunology research.

Unique Biology

PBMCs comprise a diverse population of circulating immune cells, including T lymphocytes, B lymphocytes, natural killer (NK) cells, monocytes, and dendritic cells. This cellular diversity enables simultaneous investigation of both innate and adaptive immune responses within a single experimental system.

Because PBMCs are primary human cells, they retain donor-specific genetic and immunological characteristics, making them highly valuable for studying immune heterogeneity, disease biomarkers, and personalized medicine.

PBMCs are extensively used for immune activation assays, cytokine profiling, immunophenotyping, vaccine evaluation, cancer immunotherapy research, and host-pathogen interaction studies. They also serve as one of the most important sample types for single-cell sequencing and multi-omics technologies.


Culture Considerations

Typical culture conditions include:

  • RPMI-1640 Medium
  • 10% Fetal Bovine Serum
  • L-Glutamine
  • Penicillin-Streptomycin (optional)
  • 37°C
  • 5% CO₂

PBMCs are maintained as suspension cultures and are generally used for short-term experiments. Gentle handling during isolation and culture is essential to preserve cell viability and minimize unintended immune activation.
