SOP-011: Aseptic Technique in Cell Culture
Purpose
To minimize the risk of microbial contamination during mammalian cell culture by implementing standardized aseptic handling procedures.
Scope
This SOP applies to all routine mammalian cell culture procedures performed within a certified Class II Biosafety Cabinet.
Responsibilities
The operator is responsible for:
- Following aseptic technique throughout all procedures.
- Maintaining a clean working environment.
- Preventing cross-contamination between cultures.
- Reporting suspected contamination immediately.
Required Materials
Reagents
- 70% ethanol
- Sterile culture media and reagents
Equipment
- Class II Biosafety Cabinet
- Sterile pipettes and pipette tips
- Sterile culture vessels
- Biohazard waste container
Procedure
1. Prepare the Work Area
Disinfect the biosafety cabinet work surface with 70% ethanol before beginning work.
Arrange only the materials required for the procedure inside the cabinet.
Allow sufficient space to maintain an organized workflow.
2. Prepare Materials
Inspect all media, reagents, and culture vessels before use.
Disinfect the exterior surfaces of containers before placing them inside the cabinet.
Keep sterile items sealed until required.
3. Perform Cell Culture Procedures
Work slowly and deliberately.
Open culture vessels only when necessary.
Keep lids, caps, and containers exposed for the shortest possible time.
Avoid placing hands or equipment over open sterile containers.
Use sterile pipettes or tips for every transfer.
Avoid reusing disposable sterile items.
4. Prevent Cross-Contamination
Handle only one cell line at a time whenever possible.
Change pipettes and tips between samples.
Keep culture vessels clearly labeled throughout the procedure.
5. Complete the Procedure
Close all culture vessels securely.
Remove waste promptly.
Disinfect the work surface after completing the procedure.
Return materials to their designated storage locations.
