SOP-015: Cell Culture Waste Disposal
Purpose
To ensure the safe handling, segregation, decontamination, and disposal of waste generated during mammalian cell culture procedures while minimizing risks to personnel and the environment.
Scope
This SOP applies to all biological and laboratory waste generated during routine mammalian cell culture work.
Responsibilities
The operator is responsible for:
- Segregating waste according to laboratory policy.
- Disposing of waste in designated containers.
- Preventing contamination during waste handling.
- Reporting spills or accidental exposures immediately.
Required Materials
Reagents
- Laboratory-approved disinfectant
- 70% ethanol (if required)
Equipment
- Biohazard waste bags
- Sharps container
- Liquid waste container
- Laboratory wipes
- Personal protective equipment (PPE)
Procedure
1. Segregate Waste
Separate waste into the appropriate categories:
- Biological waste
- Liquid waste
- Sharps
- Non-contaminated laboratory waste
Do not mix incompatible waste streams.
2. Dispose of Solid Biological Waste
Place contaminated disposable items, including:
- Pipette tips
- Serological pipettes
- Gloves
- Culture plates
- Culture flasks
- Tubes
into designated biohazard waste containers.
Do not overfill waste bags or containers.
3. Dispose of Liquid Waste
Collect liquid biological waste in designated containers.
Treat or dispose of liquid waste according to institutional biosafety procedures before final disposal.
4. Dispose of Sharps
Immediately discard needles, blades, broken glass, and other sharps into approved sharps containers.
Do not recap, bend, or manipulate sharps prior to disposal.
5. Clean the Work Area
After waste removal:
- Remove any visible contamination.
- Disinfect the work surface.
- Dispose of cleaning materials as appropriate.

Quality Control
Verify:
- Waste containers are correctly labeled.
- Biohazard bags are not overfilled.
- Sharps containers remain below the maximum fill level.
- Liquid waste containers are securely closed.
- Work surfaces are disinfected after completion.
