SOP-016: Operation of a CO₂ Incubator

Purpose

To ensure the proper operation and routine use of a CO₂ incubator for maintaining optimal environmental conditions for mammalian cell culture.


Scope

This SOP applies to the routine use of humidified CO₂ incubators used for mammalian cell culture.


Responsibilities

The operator is responsible for:

  • Verifying incubator operating conditions before use.
  • Minimizing disturbances to the incubator environment.
  • Maintaining cleanliness within the incubator.
  • Reporting equipment malfunctions promptly.

Required Materials

Reagents

  • Sterile distilled water (for humidity pan, if applicable)
  • Laboratory-approved disinfectant
  • 70% ethanol

Equipment

  • CO₂ incubator
  • Cell culture vessels
  • Laboratory wipes
  • Personal protective equipment (PPE)

Procedure

1. Pre-Use Inspection

Before opening the incubator, verify:

  • Temperature is within the laboratory's specified operating range.
  • CO₂ concentration is at the required set point.
  • Alarm indicators are inactive.
  • The humidity reservoir contains sufficient sterile water (if applicable).

Do not use the incubator if operational alarms are present.


2. Loading Cell Cultures

Open the incubator door only as long as necessary.

Place culture vessels on shelves with adequate spacing to allow proper air circulation.

Do not overcrowd shelves or block air vents.

Close the door securely after loading.


3. Routine Operation

Limit the frequency and duration of door openings.

Avoid unnecessary movement of culture vessels.

Monitor cultures according to laboratory schedules without prolonged exposure outside the incubator.


4. Daily Monitoring

Confirm that:

  • Temperature remains stable.
  • CO₂ concentration is within specification.
  • Humidity reservoir is adequately filled (if applicable).
  • No spills or contamination are present inside the chamber.

5. End of Use

Ensure all culture vessels are properly labeled.

Remove any spills immediately.

Return the incubator to normal operation.

  • Water reservoir is clean and adequately filled.
  • Shelves and chamber are free from contamination.
  • Incubator alarms function properly.
