SOP-017: Operation of a Laboratory Water Bath
Purpose
To ensure the safe and proper operation of a laboratory water bath for warming cell culture media, reagents, and cryopreserved cell vials while minimizing contamination risks.
Scope
This SOP applies to the routine operation of laboratory water baths used during mammalian cell culture procedures.
Responsibilities
The operator is responsible for:
- Verifying the water bath is operating correctly before use.
- Maintaining water cleanliness.
- Preventing contamination of water and samples.
- Reporting equipment malfunctions.
Required Materials
Reagents
- Distilled or deionized water
- Laboratory-approved disinfectant (for routine cleaning)

- Laboratory water bath
- Sample containers or reagent bottles
- Laboratory wipes
- Personal protective equipment (PPE)
Procedure
1. Pre-Use Inspection
Before use, verify that:
- The water level is adequate.
- The temperature has reached the required set point.
- The chamber is visibly clean.
- There are no equipment alarms or signs of malfunction.
2. Prepare the Samples
Ensure that all tubes, bottles, or containers are:
- Properly sealed.
- Clearly labeled.
- Free from external contamination.
Do not immerse open containers directly into the water bath.
3. Warm the Samples
Place the samples into the water bath carefully.
Ensure that:
- Containers remain upright.
- Water does not enter sample containers.
- Samples are removed promptly once warmed.
Avoid prolonged incubation unless specified by the protocol.
4. Remove Samples
Carefully remove the samples.
Dry the outside of each container before transferring it to the biosafety cabinet or work area.
5. Complete the Procedure
Remove any debris from the water bath.
Replace the lid after use to reduce evaporation and contamination.
