SOP-018: Operation of a Laboratory Centrifuge
Purpose
To safely and consistently operate a laboratory centrifuge for routine mammalian cell culture applications while protecting samples, personnel, and equipment.
Scope
This SOP applies to bench-top centrifuges used for processing mammalian cell suspensions, culture media, and laboratory reagents.
Responsibilities
The operator is responsible for:
- Inspecting the centrifuge before use.
- Balancing all samples correctly.
- Selecting the appropriate operating parameters.
- Reporting equipment damage or malfunction.

- Personal protective equipment (PPE)
Procedure
1. Pre-Use Inspection
Before use, verify that:
- The centrifuge is clean.
- The rotor is securely installed.
- Tubes are compatible with the selected rotor.
- No visible damage is present.
2. Prepare Samples
Ensure that:
- Tubes are properly capped.
- Samples are correctly labeled.
- Tube volumes are appropriate for the selected rotor.
3. Balance the Rotor
Arrange tubes so that opposing positions contain tubes of equal mass.
Do not operate the centrifuge with an unbalanced rotor.
4. Set Operating Parameters
Select the required:
- Speed
- Time
- Temperature (if applicable)
Verify the settings before starting the run.
5. Operate the Centrifuge
Close the centrifuge lid securely.
Start the run.
Do not attempt to open the lid while the rotor is in motion.
Allow the rotor to stop completely before opening the centrifuge.
6. Remove Samples
Carefully remove the tubes without disturbing pellets when present.
Inspect the tubes for leakage or damage before proceeding to the next step.

- Date
- Sample identification
- Centrifuge identification

- Operator initials
Quality Control
Verify:
- Rotor is securely installed.
- Samples remain properly labeled.
- No tube leakage occurred.
- No abnormal vibration or noise was observed.
- Equipment is clean after use.
Troubleshooting
