SOP-023: Cell Morphology Assessment
Purpose
To standardize the routine microscopic assessment of mammalian cell morphology for monitoring culture health, detecting abnormalities, and identifying potential contamination.
Scope
This SOP applies to routine morphological assessment of adherent and suspension mammalian cell cultures using an inverted microscope.
Responsibilities
The operator is responsible for:
- Performing routine morphological assessments.
- Recording abnormal observations.
- Identifying cultures that require further investigation.
- Reporting suspected contamination or unexpected morphological changes.
Required Materials
Equipment
- Inverted microscope
- Cell culture vessels
- Laboratory notebook or electronic record system
Procedure
1. Prepare the Culture
Remove the culture from the incubator.
Inspect the culture vessel for:
- Proper labeling
- Medium appearance
- Signs of leakage or damage
2. Examine Under the Microscope
Observe the culture using an inverted microscope.
Evaluate:
- Overall cell morphology
- Cell attachment (adherent cultures)
- Cell distribution
- Cell density
- Cell aggregation (suspension cultures)
3. Assess Cell Health
Compare the observed morphology with the expected appearance for the cell line.
Look for:
- Uniform cell shape
- Healthy cytoplasmic appearance
- Normal cell boundaries
- Appropriate cell-to-cell spacing
4. Identify Abnormal Findings
Record any abnormalities such as:
- Cell rounding
- Cell shrinkage
- Excessive debris
- Vacuolization
- Cell detachment
- Excessive aggregation
- Abnormal morphology
- Possible contamination
5. Document Findings
Record all observations before returning the culture to the incubator or proceeding with additional procedures.

Critical Control Points

Quality Control
Verify:
- Microscope is functioning properly.
- Culture vessel is clearly labeled.
- Cell morphology is consistent with previous observations.
- Records are complete and accurate.
Troubleshooting
