SOP-029: Cleaning and Maintenance of a CO₂ Incubator

Purpose​

To ensure routine cleaning and maintenance of a CO₂ incubator to minimize contamination and maintain optimal culture conditions.

Scope​

This SOP applies to all CO₂ incubators used for mammalian cell culture.

Responsibilities​

The operator is responsible for:

  • Performing routine cleaning according to laboratory schedules.
  • Maintaining incubator hygiene.
  • Reporting equipment malfunction or contamination.
  • Documenting maintenance activities.

Required Materials​

Reagents​

  • Laboratory-approved disinfectant
  • 70% ethanol
  • Sterile distilled or deionized water

Equipment​

  • Laboratory wipes
  • Personal protective equipment (PPE)
  • Biohazard waste container

Procedure​

1. Prepare the Incubator​

Remove all cell cultures, media, and accessories.

Switch the incubator to maintenance mode or power off if required by laboratory procedures.

2. Remove Internal Components​

Remove:

  • Shelves
  • Shelf supports
  • Water reservoir
  • Removable trays

Inspect all components for visible contamination.

3. Clean Interior Surfaces​

Wipe all accessible interior surfaces using a laboratory-approved disinfectant.

After the recommended contact time, remove any disinfectant residue if required.

4. Clean Removable Components​

Clean removable shelves and accessories individually.

Allow them to dry completely before reinstallation.

5. Replace Water Reservoir​

Discard remaining water.

Clean the reservoir thoroughly.

Refill with sterile distilled or deionized water according to laboratory policy.

6. Reassemble the Incubator​

Reinstall all components.

Verify that shelves are correctly positioned.

Restart the incubator if applicable.

Allow environmental conditions to stabilize before returning cultures.

Record the following:

  • Date
  • Incubator identification
  • Cleaning performed
  • Water reservoir replaced
  • Operator initials
  • Maintenance observations

Quality Control​

Verify:

  • Interior surfaces are clean.
  • Shelves are correctly installed.
  • Water reservoir contains fresh sterile water.
  • Temperature and CO₂ return to normal operating conditions.
  • Maintenance log has been completed.
