Overview

N-2 Supplement is a chemically defined, serum-free supplement designed to support the survival, proliferation, and controlled differentiation of neural stem cells (NSCs), neural progenitor cells (NPCs), and other neural precursor populations. It was one of the first defined serum replacements for neural cell culture and remains the standard supplement for neural induction and early neural development protocols.

Unlike B-27 Supplement, which is optimized for mature neurons, N-2 is primarily used during the expansion and maintenance of neural precursor cells.

Biological Role

N-2 Supplement supports neural cultures by:

  • Maintaining neural stem cell populations
  • Promoting neural progenitor expansion
  • Supporting neurosphere formation
  • Facilitating neural induction
  • Reducing variability associated with serum
  • Enabling controlled neural differentiation

Advantages

  • Chemically defined formulation
  • Serum-free
  • Excellent for NSC and NPC culture
  • Supports reproducible neural differentiation
  • Minimizes experimental variability
  • Well established in developmental neuroscience

Limitations

  • Not optimized for mature neuronal maintenance
  • Requires a suitable basal medium
  • Does not replace neurotrophic growth factors
  • Commonly used together with EGF and FGF2 for stem cell expansion
